Not if you do it in the right way.
Rather than making a big deal of it and leading up to a big question then naturally make it so that the two of you are working late one night on a project or something.
Then ask if they want to go for a drink once the project is finished.
This should let you see in a slightly more relaxed setting whether they are interested or not - if they don't even want to go for that drink then it is a sure sign that they are not!
