The most important and the most useful is probably going to have to be penicillin.
When Fleming found it it really has transformed the world with antibiotics meaning that millions of people are now alive, or have had their lives extended, than would have been the case otherwise - therefore it is hard for anything to rank above penicillin really in that capacity.
